Video summary

Moderna and Merck report positive Phase 3 melanoma results for an individualized mRNA vaccine added to Keytruda after a decade of development.

Moderna CEO Stéphane Bancel tells a16z’s Jorge Conde that Moderna and Merck’s personalized mRNA cancer treatment met its Phase 3 primary endpoint of recurrence-free survival in melanoma, and also its secondary endpoint of distant metastasis-free survival. In an earlier Phase 2 study, about 80% of patients were disease-free five years after treatment; Bancel hopes to file with regulators and make the treatment available as early as 2027. The vaccine compares tumor and healthy-cell DNA, then uses an algorithm to select up to 34 mutations and encode them in a patient-specific mRNA dose. Bancel says doses take about 42 days to produce, with a Massachusetts facility able to make tens of thousands annually. Moderna plans trials across other cancers, including lung, kidney, bladder, pancreatic and gastric cancers, while using Phase 3 patient data to improve its algorithm. He also describes future mRNA work in rare genetic liver diseases and autoimmune disease.
